Owning to the great improvement of technology and instrument, bio-imaging has obtained explosive growth, and is playing an increasingly important role in the life science research. By using imaging tools, we can show the biological phenomena in different levels, such as tissue, cell and sub-cell. Based on that, we can make qualitative and quantitative analysis on biological behavior in vivo and in vitro. With the development of optics and computing methods, the precision could achieve nanometer level in Molecular Imaging, so single molecule information in vivo could be detected. New diagnostic instruments, reagents and methods have been developed based on the combination of bio-imaging and Medical Imaging technology, promoting the disease diagnosis in cell and molecular level. It also sheds new light in the research of progression of different diseases, and then new molecular therapy strategy can be raised up.

Based on those, we are constructing an open platform with high-tech and innovation. By combining live imaging (cell and small animal level), single Molecule fluorescence imaging, high resolution confocal microscopy, ultra-high resolution fluorescence imaging (STED) technology, the most advanced sample preparation technology and data analysis, we hope to offer the best technical services to Shanghai Institute for Advanced Immunochemical Studies (SIAIS) and the whole ShanghaiTech University, also other biotech/biomedical research communities. Now, our imaging platform has been equipped with a laser scanning confocal microscope LSM710 from Zeiss, also hosted microscopy training courses regularly. In the past year, we have already provided services to SIAIS, Shanghai Advanced Research Institute (SARI), School of life Science and Technology in ShanghaiTech University and Amgen, help advancing the university's research, education and public service missions.

Instruments (under development)

l Laser Scanning confocal microscope Zeiss LSM710 upgraded to Airyscan

Equipped with 405nm/488nm/561nm/633nm laser units, LSM710 offers unrivalled confocal microscopy in cell and developmental biology. Airyscan added for higher signal-to-noise, resolution and speed.

Highlights of LSM710

(1) Oversampling with 30 percent longer sampling time per pixel for low noise

(2) Excellent contrast characteristics, even with strongly reflective samples 每 thanks to laser suppression that has been improved by 100-1000x

(3) A huge range of QUASAR detector configuration options 每 to meet your requirements

(4) The innovative grid and spectral recycling loop design ensures that not even a single valuable photon will be lost from your sample.

(5) Up to 80 percent dark noise reduction 每 thanks to the QUASAR array detector by Carl Zeiss

With Airyscan:

(1) Use a true confocal that gives you high sensitivity, improved resolution in x, y and z, and high speed in one system

(2) Increase the resolution of your imaging: with Airyscan you resolve 140 nm laterally and 400 nm axially, at 488 nm

(3) In the optional Virtual Pinhole Mode, you can decide even after the acquisition, which pinhole size best suits your application

l Inverted fluorescent microscope Olympus IX83

The fully motorized IX83 provide the ability to perform a multitude of imaging applications, from long-term time-lapse imaging and other demanding techniques to routine testing and documentation.

The fully motorized IX83 offers intuitive and ergonomic fine control over every aspect of microscope performance, with manual and automatic adjustments easily made via either the touch panel or Olympus cellSens software. The IX83 also works seamlessly with the new Olympus ZDC Z-drift compensation system, which employs a near-infrared light to provide instant, automatic focus without damaging cells. Uniquely, the Olympus ZDC unit offers both &one-shot* and &continuous* focus modes and can now even be operated stand-alone or by using the Olympus software packages. This makes it ideal for a wide range of applications, as well as flexible operation by multiple users with differing experimental requirements.

l Upright grating fluorescence microscope Zeiss Axio Imager Z2

The research microscope Axio Imager Z2 resolves details of sample from tissue sections in pathology to brain specimens in neuroscience to multi-dyed FISH specimens.

l Modular stereo microscope Zeiss SteREO Discovery. V8

The stereo microscope improved three-dimensional image perception, help acquiring crisp images throughout the whole 8:1 zoom range 每 especially in developmental biology.

l Wide zoom versatile stereo microscope Olympus SZX16

The SZX16 motorized focus drive makes digital documentation with extended focal imaging efficient and fully automatic.

l Super-resolution in vivo multiphoton fluorescence small animal imaging system (self-assembled)

Construction of the system can be used for small dynamic ultra-high resolution and multi-color fluorescence imaging of living. The device can be used to study various physiological and pathological models such as nerve, stem cell, growth, inflammation, cancer, skin diseases at the animal level.

l Rotary confocal microscope Perkin Elmer UltraVIEW VoX (co-build with PE)

The UltraVIEWVoX 3D spinning disk live cell imaging system offers acquisition of detailed cellular structure, also the 3D analysis provides great context to cellular changes and more accurate results.

l Inverted fluorescence microscope Zeiss Axio Observer A1

Axio Observer is a platform for gentle imaging of living cells, with the software controlled incubation modules, it regulates temperature, CO2 and oxygen concentration to create suitable environment for live cell experiments.
