主要研究内容
病毒和细菌都是非常重要的病原体,它们可引发多种严重传染病。就病毒而言,有史以来关于病毒性疾病的记载最早可追溯至5700年前,在古埃及的象形文字中对瘫痪的小儿麻痹症患者的描述。在现代社会中,新冠肺炎、“非典”、艾滋病、寨卡综合征和流感等一系列由病毒引发的重大传染病严重危害人类健康,并产生一系列经济问题和社会问题。在细菌方面,结核分枝杆菌是细菌类病原体的典型代表。全世界约有四分之一的人口被结核杆菌感染,而因结核病致死的人数甚至已超过了艾滋病。
研究组长期从事上述重要病原体引发的重大疾病的研究,并针对性的开展全新药物开发工作。主要涉及的研究方向包括:1、重要病原体的关键药物靶点的结构和功能研究;2、病原体入侵宿主过程中二者之间的精确相互作用和分子机制,特别关注宿主免疫系统与病原体之间“抑制与拮抗”的复杂斗争过程;3、针对关键药物靶点开展合理药物设计、先导药物的高通量筛选以及临床前评价。
研究成果展示 杨海涛教授担任了国家重点研发计划总体专家组专家、国家“重大新药创制”科技重大专项评审专家、法国国家科研署基金外审专家、波兰国家科学基金外审专家、香港特区政府研究资助局评审专家、中国博士后科学基金特别资助专项评审专家、教育部全国博士学位论文抽检评议专家、国家重点基础研究发展计划(973计划)青年项目首席科学家;担任了Nature、The Lancet Infectious Diseases、Nature Chemical Biology、Nature Communications、Science Advances、Angewandte Chemie、Chemical Science、Journal of Medicinal Chemistry、ACS Catalysis、The Lancet's EBioMedicine和Journal of Antimicrobial Chemotherapy等十余种国际知名杂志的审稿人;担任了中国期刊Cell Research、Protein & Cell、Science Bulletin和Journal of Molecular Cell Biology的审稿人。获得了中国产学研合作促进奖、中国侨界贡献奖、科学中国人年度人物、谈家桢生命科学创新奖、树兰青年医学奖、Promega Award for Biochemistry和药明康德生命化学杰出成就奖等奖项。近期研究成果包括: 1、在结核杆菌关键药物靶点的结构和药物发现的研究中取得进展,论文发表于Cell、Science等杂志,并入选2019年度“中国高校十大科技进展”和2019年度“中国生命科学十大进展”。 2、在新型冠状病毒关键药物靶点的结构和药物开发的研究中取得进展,相关研究论文先后在Nature、Science(封面论文)、Cell和Nature Structural & Molecular Biology等杂志发表,并入选2020年度富林沃斯(Falling Walls)生命科学领域十项进展、2020年度“中国生命科学十大进展”。除此之外,相关研究亦与我国其他抗疫成果一起集体入选“2020年度中国科学十大进展”。
